HONOLULU, HI— Panadol Advance (paracetamol; fast-absorbing) 1,000mg tablets were more efficacious and demonstrated faster onset and longer duration of pain relief compared with Panadol Advance 500mg, standard paracetamol 650mg, and placebo, reported Yong Yue, MD, from Jean Brown Research, Salt Lake City, UT at the American Pain Society’s 31st Annual Scientific Meeting.
Panadol Advance is a fast absorbing formulation of paracetamol. Dr. Yue and colleagues conducted two single center, single dose, randomized, placebo-controlled, double-blind, double-dummy, parallel group studies to evaluate the efficacy and time to onset of pain relief over a period of six hours of Panadol Advance 1,000mg versus Panadol Advance 500mg or standard paracetamol 650mg versus placebo in patients with post-surgical dental pain following third molar extraction.
Study I assessed single doses of Panadol Advance 1,000mg, 500mg, or placebo in 301 patients. Study II evaluated single doses of Panadol Advance 1,000mg, paracetamol 650mg, and placebo in 400 patients. Patients in both studies had similar baseline pain intensity scores.
Results showed that Panadol Advance 1,000mg demonstrated significantly greater pain relief compared with Panadol Advance 500mg, paracetamol 650mg, and placebo, as measured by Sum of Pain Relief and Pain Intensity Difference (SPRID) and other endpoints for up to six hours post-dose. SPRID values were significantly greater for Panadol Advance 1,000mg vs Panadol Advance 500mg, paracetamol 650mg, and placebo through 6 hours in both studies. Panadol Advance 500mg and paracetamol 650mg also demonstrated greater efficacy over placebo, but had inferior duration of action compared with Panadol Advance 1,000mg.
Onset of pain relief (first perceptible pain relief) was observed at 15 minutes in patients who received Panadol Advance 1,000mg, a 32% and 25% reduction in time than that of Panadol Advance 500mg (22 minutes) and paracetamol 650mg(20 minutes), respectively. Pain relief scores (PRS) were significantly greater for Panadol Advance 1,000mg over placebo from 15 to 240 minutes post dose in Study I, and through 6 hours in Study II, and over Panadol Advance 500mg from 45 to 300 minutes post dose in Study I and from 15 to 300 minutes (except 45, 60, and 90min) post dose in Study II.
Dr. Yue noted that all treatments were well tolerated and went on to conclude that Panadol Advance 1,000mg is an effective option for pain relief.
